内质网 (endoplasmic reticulum,ER) 是细胞 中蛋白质、类固醇、脂类的合成场所,对于细胞的 活动和生存是不可缺少的。多种因素使 ER 稳态被 打破,发生错误折叠和未折叠蛋白反应 (unfolded protein response,UPR),在 ER 腔内聚集以及引 起钙离子平衡紊乱,称为内质网应激 (endoplasmic reticulum stress,ERS)。葡萄糖调节蛋白 78 (glucose regulated protein 78,GRP78) 作为一种分子 伴侣,在蛋白质的折叠、转运过程和 ERS 反应中发 挥重要作用。近年发现,GRP78在泌尿系肿瘤中过 表达,可能与泌尿系肿瘤,如前列腺癌、肾细胞 癌、膀胱癌的恶性生物学行为,如增殖、凋亡、侵 袭等密切相关[1-2] ,本文对 GRP78表达在泌尿系肿瘤 的研究进展作一综述。

GRP78 在泌尿系肿瘤,如前列腺癌、膀胱癌、 肾癌呈高表达,与肿瘤增殖、转移、耐药等恶性生 物学行为密切相关,提示其可能是一个良好的生物 学标志物和治疗靶点。GRP78联合新型纳米颗粒给 药系统的初步成功,也为抗肿瘤治疗提供了新的方 向和希望。但目前关于 GRP78在泌尿系肿瘤的相关 研究大多数为基础研究,具体作用机制需要进一步 探讨,还需要大规模、多中心、随机对照研究证实 其临床有效性。相信随着研究的深入,GRP78可能 成为泌尿系肿瘤一个有效的治疗靶点和预后标志物。
